Biotech lab assignment Monday 4/21 (due tomorrow Tuesday):
 
You may find useful the CH 6 glossary on E16-17.
 
Pg E-3: “What do you already know” questions 1-4. Use section 19.6 (bacteria) and sections 3.5 and 3.6 (proteins) of the Honors biology text to assist you in answering the questions. Do these questions first, and make sure you get them done in class.
 
Read E4-E8 and answer the “consider” questions. Before each answer, write, “consider” followed by the page number, to identify the question. EX: “consider E4”.
 
Read the top paragraph on E9, and then on the same sheet of paper you have been working on, complete “before the lab” questions 1-2.

Have this done before coming to class Monday 4/21:

Read Lab 6a pg. 87-89.  Review, but don't answer "what do you already know"?  Remember early in the year when we discussed protein structures (primary, secondary, tertiary, quaternary), such as hemoglobin, which forms a "bird's nest" to carry oxygen.

Read pg. 90-95.  Answer no questions.

Skim pg. 96-103 to be familiar with lab 6.

In class, turn in your homework first before sitting down. Sit down every day with ONLY your ALL of your biotech labs, a new sheet of paper, and 2 working pencils and an eraser. Let me know if there are not 2 copies of the AMGEN student manual on your desk. Each should be facing opposite of each other so everyone can read from it. ALL notebooks and other supplies should be AWAY in your BAG, not on the floor or on the counters. We will continue to work in groups of three.
DO NOT HAVE ANY FOOD OR DRINKS OUT. They must be IN YOUR BAG, not on a counter.
 
You do not need to write out and answer the “Stop and Think” Questions. Just, stop, read them, and think about them for a moment.

1)      Complete the ENTIRE #3 from “Before the Lab” on pg. C44 for Lab 5a. You are able to do this ENTIRE problem even before viewing your plates after 24 hours of incubation. Be sure to use table 1 and table 2 from RM5 (says in top right corner) handout you received in class today.
2)      Don’t worry if your plates will incubate for more than 24 hours. I will refrigerate them.
3)      Chapter 5a Questions 1-6 are not assigned now, but will be assigned AFTER you view your plates on Thursday.
Reminder - Genetcis Project Due Date
Due on MONDAY 4/28.  USB is due as well as printed copy of slides, one slide per page (or "sub-slide" such as "2a" if you have them).

2)      Due to time constraints, the actual lab for 2a was performed by the teacher. Some of the materials required for 4a will come from the 2a lab performed by the teacher.
3)      Lab 4a: Read C21-C29. Review C30-C32. No questions tonight from lab 4a. Be ready for the lab tomorrow.

3)      Start Lab 2A (Lab 2A, not Lab 2). No need to answer the questions where it says “Consider”. Just consider them, and then move on.
4)      From the Amgen Student Guide, read pgs. C1-C8
5)      Review C9-C11 (do not do questions, just review them)
6)      On a separate sheet of paper: Read C12-C13 and do “Before the Lab” questions 1-4 on page C13. Do these on a sheet of paper SEPARATE from your gel lab today in class.

Directions 
General Info.
Use lined paper for these biotech labs. Staple each day’s work together for this three-week lab.  Put your name, date, and period # top right. Keep all of the different days for this three-week lab paperclipped together, in order. Bring them to your seat every day. Do not lose them, as you will be graded on the completeness of your entire lab at the end of the three weeks.
 
The Student Guide for the Amgen Biotech Experience is on this Group Fusion site as both a link and a downloadable PDF (go to "Files", and download the file called "student.Amgen.pdf".  This is your guide for this entire lab experience. Do not remove the hardcopy of this book from the classroom. It is needed for other classes. 
 
Classwork After Quiz Monday/Homework if you don’t finish in class:
In the Amgen Student Guide, read pgs. A-1, A-2, A-5, A-6, A-12, A-13. If you did not finish in class, do so tonight. The homework MUST be completed prior to lab on Tuesday 4/1.
 
Homework tonight- required for lab on Tuesday 4/1
1)      From the Amgen Student Guide, read pg. A-17 and answer questions 1-2 at the bottom (15 min. max). Use a separate sheet of lined paper always for these biotech labs. Title appropriately. At this point, you should be able to do these from memory; however you may use your text if necessary.  For #1, part 2, you only need to provide three examples.  For #2, there is no need to “be as detailed as possible” as the guide says. You do, however, need to answer the questions to the extent that we have learned them. 
2)      Read pg. A-18 (one min.), and then watch the video on micro-pipetting using this link:
3)      Following the video, on the same sheet of paper as the answers from page A-17, answer question #1 (only) from page A-19, “Before the Lab”. Title appropriately.  For question #2, do not answer, but take a minute or two to skim through pages A-19 through A-21 to familiarize yourself with tomorrow’s lab.
 
If you have time Monday night, do this also. Otherwise, must be completed by Tuesday Night:
1)      Read A-7 through A-11., and A-14.
